外源性孕激素补充影响胚胎着床期免疫的实验研究

【摘要】: 目的:观察比较孕鼠着床期子宫内膜中免疫调节蛋白孕酮诱导的封闭因子(progesterone-induced blocking factor,PIBF)、雌激素受体(estrogen receptor-a,ER-a)、孕激素受体(progesterone receptor,PR)的表达以及血清雌二醇(estradiol,E_2)及孕酮(progesterone,P)含量差异,探讨外源性孕激素补充对胚胎着床期免疫的影响及其影响途径。 方法:昆明小鼠随机分为地屈孕酮组(从孕1天起每日灌胃2mg地屈孕酮)、黄体酮组(从孕1天起颈背部皮下注射2mg黄体酮)和对照组,分别于孕4、5、6天收集子宫及血清;C57BL/6雌鼠随机分为同基因交配组(与C57BL/6雄鼠合笼交配)和异基因组(与BALB/c雄鼠合笼交配),在孕6天收集子宫及血清。免疫组化法检测各组子宫内膜PIBF、ER-a、PR表达,放射免疫法检测血清E_2及P含量,对比分析各组间差异。 结果:(1)异基因交配组(C57BL/6×BALB/c)较同基因交配组(C57BL/6×C57BL/6)子宫内膜PIBF表达明显增高(p<0.05),血清E_2、P含量和子宫内膜ER-a、PR表达均显著升高(p<0.05)。(2)外源性孕激素补充(包括天然黄体酮和地屈孕酮)的孕鼠其子宫内膜PIBF表达明显增高(p<0.05),地屈孕酮组较黄体酮组PIBF表达更高,差异有显著性(p<0.05)。黄体酮组血清P含量显著升高(p<0.01),ER-a和PR表达下降(p<0.05)。地屈孕酮组血清E_2、P含量无变化(p>0.05),但子宫内膜ER-a、PR表达显著升高(p<0.05)。 结论:PIBF的表达需要异基因胚胎抗原的刺激;地屈孕酮和黄体酮均上调PIBF表达,,地屈孕酮的作用强于黄体酮,且前者上调而后者下调子宫内膜ER-a、PR的表达。
[Abstract]:Objective: to observe and compare the blocking factor (progesterone-induced blocking factor,PIBF), estrogen receptor (estrogen receptor-a,ER-a) and progesterone receptor (progesterone receptor, induced by progesterone in the uterus of pregnant rats during implantation. The expression of PR) and the difference of serum estrogen (estradiol,E_2) and progesterone (progesterone,P) levels were studied to explore the effect of exogenous progesterone supplementation on embryo implantation immunity.
